Title and Brief report of the work (max 300 words)

Site-directed mutagenesis of L1 gene

Introduction

Tobamo viruses are RNA viruses having positive-sense, single-stranded RNA genome, encapsulated into rigid, rod-shaped particles. Tobamo viruses are contact transmitted and can be classified into pathotypes according to their ability to overcome host resistance. Capsicum species has been characterized to express a resistant protein (encoded by L gene) in response to tobamo virus infection. The resistance against different tobamo viral pathotypes (P0 to P1,2,3) is conferred by four different L gene alleles (L1 to L4) .

Objective:

To convert L1 to thermo resistant L1a allele, four amino acids were selected for Site-directed mutagenesis.

Materials and methods:

Substitution mutations were introduced by site-directed mutagenesis using the Quick-Change Kit (Stratagene). Plasmid comprising L1 gene (pEAQ-HT-DEST1-L1) was used as a template for generating a mutation in L1 gene.

Results:

Plasmid comprising L1 gene (pEAQ-HT-DEST1-L1) was used as a template for the PCR reaction with L1 296 SC F and L1 296 SC R primers.
