Title: Role of Tandem Zinc Finger proteins of Arabidopsis thaliana in plant stress responses. In the present study, it is observed that Tandem Zinc Finger proteins (TZF7-TZF11) localized to cytoplasmic foci and also in the nucleus and/or cytoplasm. Colocalization analyses reveals the proteins’ association with the various P-body, siRNA body and stress granule markers. Interestingly, “stress granules” labelled with the SG markers, PAB2 and PAB8, are observed to be formed even in the absence of stress, when co-expressed with the TZFs. Expression of TZF (TZF7-TZF11) genes is observed to be PAMP (Pathogen-Associated Molecular Pattern) inducible and also the TZFs are observed to be phosphorylated by PAMP-responsive mitogen-activated protein kinases (MAPKs). Also changes in the speckled localization of the TZFs are observed upon PAMP elicitation (until 1 hr). Mutant analysis indicates that the quadruple and the penta mutants show hypersensitive responses to abscisic acid (ABA) induced root growth inhibition. tzf10 single insertion mutant show severe yellowing of leaves in the presence of ABA, possibly depicting its dominant role in ABA dependent growth responses. These results suggest the involvement of the TZFs in post-transcriptional regulation and thereby, regulating plant responses to ABA. |
