Title and Brief report of the work (max 300 words)

I had participated in ongoing gene expression analysis experiments to evaluate different candidate genes to be used as housekeeping genes on qPCR, and DNA quantification in different tissue samples of sorghum crop. In parallel, I studied various literature on the genomics approaches planned in the BIOSORG project and presented a journal club on the concept of "Genome wide association study (GWAS), RNA seq experiments and real time PCR expression analysis". Due to delays in the timelines of the BIOSORG project, the academic supervisor proposed me an additional training and research activities in the framework of ClimBar, an international project aimed to develop “an integrated approach to evaluate and utilize genetic diversity for breeding climate-resilient barley”. In this context, I get involved in dissecting natural genetic variation for culm diameter and culm wall thickness in the ClimBar panel under field conditions with collection of green and straw internodes at dough and harvesting stage respectively. I successfully completed the task for sectioning of the dried culm samples of all the barley accessions to measure their diameter. Besides, I learned to use digital image analysis software to obtain quantitative measurements from culm section images. Finally the data were subjected to analysis of variance and corrected means of each accession were obtained for further analyses using an appropriate linear statistical model implemented in statistical package R. Research outcomes includes, Hands-on experience in the use of DNA sequence databases, sequence retrieval from public databases, sequence analysis programs, finding local alignments between sequences, discovery of evolutionary relationships using sequences and the FASTA and BLAST methods for database searches etc., use of image analysis software ImageJ and statistical analysis software like R, SAS etc. Well aware about the techniques on pre and post-harvest culms phenotyping in the Barley crop.
