Gene Expression Analysis for biosynthesis and catabolism of Glucosinolates in Rocket (Diplotaxis tenufolia.L) under abiotic stress

As rocket (Diplotaxis tenufolia L.) is one of those leafy vegetables which are largely preferred consumed in western diets for its rich peppery taste and pungent aromas. It has been reported to contain high levels of vitamin C, glucosinolates, flavonols, and phenolics. Among these, glucosinolates are one of the important nutritive content that is responsible for its sharp and bitter-tasting flavor. Glucosinolates and many of its hydrolysis products are reported to have anticancerous, antibacterial, antifungal properties as well as other health promoting compounds. The present study was conducted in order to increase glucosinolates in these leafy vegetables so that, it could be a dietry supplement for meat eaters or those less dependent on veggies. In this experiment, rocket plant was grown in the greenhouse and heat and salt stress treatment was given to the plant after a month. Physiological and biochemical analysis was done which was further followed by expression profiling and full length isolation of some interested genes. Content of chlorophyll and chlorophyll fluorescence were measured. Glucose and glucosinolate content of the rocket leaves and roots were also measured through D-Glucose Assay. Expression pattern of glucosinolate biosynthesis and catabolism was confirmed by designing primers for the specific genes involved in the glucosinolate biosynthetic and catabolic pathway. Expression analysis was performed through Real Time PCR using ΔΔCT method. The experiment was further followed by the full length isolation of two of the most interested genes using RACE method. The paper is under publication so the results have not been discussed here.
