Title and Brief report of the work (max 300 words)

1. Master thesis: Genetic engineering to confer resistance against Sri Lankan cassava mosaic virus (SLCMV) using tobacco as a model plant. 

Research description: Sri Lankan cassava mosaic virus (SLCMV) is the principal cause of cassava mosaic disease in the Indian subcontinent. To gain resistance against the virus, the coat protein (CP) gene of SLCMV-Adivaram isolate was cloned in either sense or antisense orientation under 35S promoter and transgenic Nicotiana benthamiana plants were raised through Agrobacterium-mediated transformation. A total of eight T1 transgenic lines, four harbouring the sense-CP construct and four harbouring the antisense-CP construct were challenged with agro-infectious clones of SLCMV DNA-A and DNA-B. We found that three out of four sense-CP transgenic lines and all four antisense-CP transgenic lines gave on an average 65.67% and 67.25% delay in symptom appearance until 20 days post-inoculation, respectively. Moreover, the viral titre was lower in the challenged transgenic lines compared to the wild-type plants, as confirmed by qPCR and dot blot analysis. Furthermore, we have detected viral specific siRNAs (vsiRNAs) in resistance transgenic lines through small RNA Northern blot analysis, suggesting the potential involvement of the RNA interference pathway in providing the observed resistance against the virus. 

 

2. Exogenous application of double-stranded RNA molecules on tomato leaves, its systemic trafficking and movement of silencing signals to importance agricultural pests   viz. aphids, whiteflies and mites.

Project Description: Exogenously applied double-stranded RNA (dsRNA) molecules onto tomato leaves, moved rapidly from local to systemic leaves and were uptaken by agricultural pests namely aphids, whiteflies and mites. Four small interfering RNAs, deriving from the applied dsRNA, were molecularly detected in plants, aphids and mites but not in whiteflies.
