Title and Brief report of the work (max 300 words) | Experiment performed dsRNA mediated protection against the Tomato leave curl Gujarat virus and Cucumber mosaic virus. Host : Nicotiana benthamiana Host : tomato Result With each host plant, experiments are repeated for more than 3 times and final result, in case of Nicotiana benthamiana result was 2 to 3 days delay in viral infection in dsRNA treated plant as compared to control treatment. In tomato, dsRNAs for AC1/AC4, AV1/AV2, 1/AC4- /AV2 and CMV_2b-ToLCGV_AV1/AV2 conferred 45%, 60%, 50% and 55% resistance, respectively. Experiments with Nicotiana tabacum showed that the dsRNA construct CMV_2b-ToLCGV_AV1/AV2 conferred 33.3% resistance against CMV, while CMV_2b provided 40% resistance. This is the first case where RNA-based vaccination is functional for a geminivirus and where a single dsRNA molecule could induce resistance against two tomato infecting viruses, namely an RNA (CMV) and a DNA (ToLCGV) plant virus. Therefore, the production of single dsRNA molecules containing the desired targets could provide control against different viruses infecting the crop of interest. We also performed experimental study on systemic transport dsRNA in tomato. |
